Eli Lilly will present new clinical data from its cardiometabolic portfolio at the 62nd Annual Meeting of the European Association for the Study of Diabetes (EASD), including Phase 3 results for investigational retatrutide, Phase 3 findings for orforglipron, cardiovascular data for tirzepatide, and Phase 2 results for the investigational eloraTZP combination.
Retatrutide Shows Weight and A1C Improvements in Type 2 Diabetes
Retatrutide is an investigational once-weekly triple hormone receptor agonist that activates receptors for glucose-dependent insulinotropic polypeptide (GIP), glucagon-like peptide-1 (GLP-1), and glucagon. Lilly will present Phase 3 results from the TRIUMPH-2 study in adults with obesity or overweight and type 2 diabetes.
At 80 weeks, participants receiving retatrutide lost up to an average of 49.6 lb (22.5 kg), equivalent to 20.8% of body weight. Average A1C reductions reached 1.6%. These findings will be presented during an EASD-sponsored retatrutide symposium on September 30.
The broader Phase 3 program is evaluating retatrutide in obesity and overweight with weight-related medical conditions, type 2 diabetes, knee osteoarthritis, moderate-to-severe obstructive sleep apnea, chronic low back pain, cardiovascular and renal outcomes, and metabolic dysfunction-associated steatotic liver disease. Retatrutide remains investigational and is available only to participants in Lilly clinical trials.
Orforglipron Provides Phase 3 Cardiovascular Data
Foundayo (orforglipron) is a once-daily oral, non-peptide GLP-1 receptor agonist. Lilly will present Phase 3 results from the ACHIEVE type 2 diabetes program, including findings from ACHIEVE-4 in adults with type 2 diabetes and obesity or overweight who had increased cardiovascular risk.
In ACHIEVE-4, orforglipron demonstrated non-inferiority to insulin glargine for major adverse cardiovascular event (MACE) endpoints. The reported hazard ratio was 0.84 (95.0% CI, 0.59-1.20) for MACE-4 and 0.77 (95.0% CI, 0.52-1.13) for MACE-3.
Additional presentations will report time-to-goal analyses comparing orforglipron with dapagliflozin and oral semaglutide, as well as physical and psychosocial functioning outcomes from the ATTAIN-1 obesity study.
Tirzepatide Data Expand Cardiometabolic Evidence
Mounjaro (tirzepatide) is a dual GIP and GLP-1 receptor agonist. Lilly will present SURPASS-CVOT findings in adults with type 2 diabetes and established cardiovascular disease.
Tirzepatide demonstrated non-inferiority versus dulaglutide, with an 8% lower observed risk of MACE-3 events and a hazard ratio of 0.92 (95.3% CI, 0.83-1.01). The study also showed greater reductions in A1C and body weight with tirzepatide.
Other presentations will examine beta-cell function and insulin sensitivity in early type 2 diabetes and glycemic control during Ramadan fasting.
EloraTZP Combines Amylin and GIP/GLP-1 Activity
EloraTZP combines eloralintide, an investigational selective amylin receptor agonist, with tirzepatide. Lilly will present Phase 2 results in adults with obesity or overweight and type 2 diabetes.
Earlier Phase 1 results showed that eloralintide 3 mg added to tirzepatide 5 mg produced 17% weight loss over 16 weeks, compared with 10% with tirzepatide 5 mg alone.
Eloralintide is being evaluated as a once-weekly selective amylin receptor agonist in Phase 3 studies for obesity and for obesity or overweight with or without type 2 diabetes.
EASD 2026 Will Feature Multiple Cardiometabolic Approaches
Lilly’s presentations at EASD 2026, scheduled for September 28 to October 2 in Milan, will cover several approaches to treating obesity and type 2 diabetes, including triple hormone receptor agonism, oral GLP-1 receptor activation, dual GIP/GLP-1 receptor agonism, and combination therapy involving amylin receptor activation.
The company will also host an industry symposium on September 28 covering treatment of obesity and type 2 diabetes from earlier stages of disease, oral therapies, and emerging mechanisms.
