BeOne and Revolution Medicines will develop RAS(ON) inhibitor combinations and expand four RAS-targeted therapies across selected Asian markets.

BeOne Medicines and Revolution Medicines have entered a multi-part collaboration to evaluate combinations of BeOne oncology assets with four clinical-stage RAS(ON) inhibitors and to expand access to the Revolution Medicines portfolio across selected Asian markets. The agreement covers daraxonrasib, zoldonrasib, elironrasib and RMC-5127, targeting cancers driven by oncogenic RAS signaling.

Collaboration Targets RAS-Addicted Cancers

The clinical collaboration will investigate combinations of selected BeOne therapies with Revolution Medicines’ RAS(ON) inhibitor portfolio. The initial planned combinations include BeOne’s MTA-cooperative PRMT5 inhibitor BGB-58067 and its EGFR × MET × MET trispecific antibody BG-T187, each combined with either daraxonrasib or zoldonrasib.

RAS alterations are among the most common oncogenic drivers in human cancers, but developing effective therapies has been challenging because RAS proteins have historically been difficult to target directly. RAS(ON) inhibitors seek to block the active form of RAS and suppress signaling that supports tumor growth and survival. The four assets in the agreement target different RAS variants, potentially enabling a broader approach across molecularly defined cancers.

Four Clinical-Stage RAS(ON) Inhibitors Included

Revolution Medicines’ portfolio includes daraxonrasib, a RAS(ON) multi-selective inhibitor; zoldonrasib, which selectively targets RAS(ON) G12D; elironrasib, a RAS(ON) G12C-selective inhibitor; and RMC-5127, a RAS(ON) G12V-selective inhibitor.

The companies did not disclose clinical efficacy or safety results from the planned combination studies. Instead, the agreement establishes a framework for evaluating whether complementary mechanisms can improve treatment options for patients whose tumors remain dependent on RAS signaling.

 Regional Rights Expand Development and Commercial Reach

Revolution Medicines granted BeOne exclusive rights to develop and commercialize, or solely commercialize depending on the market, the four RAS(ON) inhibitors in selected Asian territories, primarily across Greater China and Southeast Asia, where BeOne, formerly BeiGene, has an established commercial and development infrastructure. Japan and South Korea remain outside the licensed territory, with Revolution Medicines retaining development and commercial rights in those markets and elsewhere outside the partnered region.

Under the agreement, Revolution Medicines is eligible for development and sales milestone payments as well as tiered royalties on net sales in the licensed territories.

A key component of the transaction is BeOne’s commitment to fund and conduct a global registrational Phase 3 study for one of the four RAS(ON) inhibitors. Revolution Medicines will continue advancing its broader portfolio through global registrational studies.

Development Strategy Moves Toward Combination and Global Expansion

John V. Oyler, co-founder, chairman and CEO of BeOne, said the collaboration will allow the companies to evaluate combinations between their respective oncology pipelines while using BeOne’s development and commercial infrastructure to expand access in Asia.

Revolution Medicines CEO and chairman Mark A. Goldsmith said the partnership will broaden the reach of its RAS(ON) inhibitors while supporting exploration of new combination strategies.

The agreement does not yet establish clinical benefit for any of the proposed combinations. Its significance lies in combining a broad RAS(ON) inhibitor portfolio with complementary oncology assets and committing resources to late-stage development, including a global Phase 3 program. Future clinical data will determine whether these combinations can translate the biological rationale into improved outcomes for patients with RAS-driven cancers.
