Kura Oncology spinoff Caspian Therapeutics raises $50 million to advance KO-7246, a menin inhibitor targeting β-cell regeneration in diabetes.

Caspian Therapeutics has launched with $50 million in financing to advance KO-7246, an oral menin inhibitor that expanded pancreatic β-cell mass and increased endogenous insulin production in preclinical models of Type 1 and Type 2 diabetes. The company plans to take KO-7246 through IND-enabling studies and initial clinical proof of concept while also advancing a second menin inhibitor candidate for diabetes and broader cardiometabolic diseases.
Menin Inhibition Targets β-Cell Loss
Kura Oncology established Caspian around KO-7246 and related intellectual property and development expertise. BVF Partners led the $50 million financing, with participation from the T1D Fund, Invus, Montanova, Eli Lilly and Company, Kura Oncology, members of Kura’s leadership team, and several successful biotech entrepreneurs.
Caspian is pursuing menin inhibition as a potential way to address a central feature of diabetes: declining pancreatic β-cell number and function. Menin acts as a regulatory scaffold and restrains β-cell proliferation. Blocking menin may therefore increase functional β-cell mass and restore the body’s ability to produce insulin.
In preclinical studies, menin inhibition selectively stimulated β-cell proliferation, increased β-cell mass, boosted endogenous insulin production, and improved glycemic control across multiple animal models of Type 1 and Type 2 diabetes.
The effects persisted after treatment stopped, suggesting that the intervention may produce a durable increase in functional β-cell capacity rather than only a transient glucose-lowering effect.
Human pancreatic islet microtissues and organoid models provided additional translational support. KO-7246 increased β-cell proliferation and function without detectable proliferation of non-β cells in these systems.
KO-7246 Moves Toward the Clinic
KO-7246 is a highly selective, orally bioavailable small-molecule menin inhibitor optimized for chronic use in metabolic disease. Caspian plans to advance the compound through IND-enabling development and submit an investigational new drug application as soon as practicable.
The initial clinical program is expected to assess safety, tolerability, pharmacokinetics, and pharmacodynamics, alongside measures of β-cell function and endogenous insulin production. No human efficacy or safety data have yet been reported.
Additional preclinical findings will be presented at the 62nd European Association for the Study of Diabetes Annual Meeting on September 29, 2026. The oral presentation, titled “Next-generation menin inhibitor KO-7246 regenerates beta cell mass in diabetic rodent and human islet models,” will be delivered by Kura Chief Scientific Officer Francis Burrows, Ph.D.
Caspian Builds a Broader Menin Inhibitor Pipeline
The financing extends beyond KO-7246. Caspian plans to use the proceeds to advance a second menin inhibitor development candidate for diabetes and cardiometabolic diseases, giving the company a broader pipeline around menin biology.
The strategy builds on Kura’s experience in menin inhibitor discovery and development through its oncology programs, including ziftomenib. Caspian will operate with dedicated leadership and external capital while accessing Kura’s research, development, and corporate capabilities under an intercompany services agreement.
Kura will retain approximately half of Caspian on a fully diluted basis and maintain board representation. This structure allows Kura to preserve substantial economic exposure to the metabolic pipeline while remaining focused on oncology.
New Leadership and Next Milestones
Robert Spencer, Ph.D., has been appointed Caspian’s President and Chief Operating Officer. He brings more than two decades of biotechnology and pharmaceutical experience across drug development, regulatory strategy, and corporate leadership, including roles at Neurommune Therapeutics, Cara Therapeutics, and Merck Research Laboratories.
The immediate development priority is to move KO-7246 through IND-enabling studies and into first-in-human testing. The clinical program will need to establish whether the β-cell expansion and increased endogenous insulin production observed in preclinical systems translate into measurable biological effects in people with diabetes.
The $50 million financing is expected to fund KO-7246 through initial clinical proof of concept while supporting development of the second menin inhibitor candidate. The September EASD presentation will provide the next detailed look at the preclinical evidence underlying Caspian’s approach.
