Nxera Pharma announced on November 4, 2026, that its subsidiary, Nxera Pharma UK Ltd, had entered into an exclusive global licensing agreement with AbbVie for NXE0033744, an investigational small-molecule EP4 receptor agonist being developed for inflammatory bowel disease (IBD).

Under the agreement, AbbVie receives worldwide rights to develop and commercialize the candidate in IBD. Nxera will receive US$100 million upfront and is eligible for up to US$1.37 billion in additional development and commercial milestone payments, alongside tiered royalties on net sales. The transaction remains subject to customary antitrust clearances, including under the US Hart-Scott-Rodino Antitrust Improvements Act.

The agreement transfers global development and commercialization responsibilities to AbbVie while allowing Nxera to retain potential long-term financial participation through milestone payments and royalties.

Mechanism of Action

NXE0033744 is an investigational oral, small-molecule EP4 receptor agonist discovered using Nxera’s proprietary NxWave™ drug discovery platform. The company describes it as a potential first-in-class candidate for IBD.

The candidate is designed to selectively activate EP4 receptors locally in the gastrointestinal tract, aiming to produce anti-inflammatory effects while supporting epithelial barrier homeostasis and mucosal healing. Its gut-targeted design is intended to minimize systemic exposure, potentially differentiating its pharmacological approach from existing IBD treatments.

This strategy addresses two important aspects of IBD pathology: persistent intestinal inflammation and disruption of the epithelial barrier. However, whether local EP4 activation can translate into meaningful improvements in clinical outcomes remains to be established.

Clinical Development

Nxera has completed a first-in-human Phase 1 programme registered under ISRCTN70080074. The programme comprised single- and multiple-ascending-dose studies, a cohort of patients with ulcerative colitis and an indomethacin-challenge cohort.

According to Nxera, the results supported a gut-restricted pharmacological profile and demonstrated target engagement through a reduction in indomethacin-induced intestinal permeability. The company also reported that no adverse events of concern were observed.

The reduction in intestinal permeability following indomethacin challenge provides an early pharmacodynamic signal consistent with the candidate’s intended mechanism. However, these findings do not establish therapeutic efficacy in IBD or demonstrate improvements in clinical remission or mucosal healing.

The ISRCTN registry does not currently provide the full Phase 1 study details or results, as their publication has been deferred.

IBD principally includes ulcerative colitis and Crohn’s disease, both chronic, relapsing inflammatory conditions. Despite advances in treatment, many patients do not achieve or maintain adequate and durable remission or mucosal healing. NXE0033744’s locally targeted mechanism represents an alternative therapeutic approach, although its potential to address this unmet need will depend on subsequent clinical evidence.

Development and Commercial Implications

AbbVie’s exclusive global rights position the company to advance NXE0033744 through further clinical development and potential commercialization. Nxera cited AbbVie’s expertise in immunological diseases and global clinical development as important capabilities for advancing the programme.

Nxera President and CEO Christopher Cargill said the agreement reflects confidence in the NxWave platform and the candidate’s locally targeted biological approach. He also highlighted AbbVie’s existing neuroscience discovery collaboration with Nxera, describing the latest agreement as further validation of the platform’s potential.

For AbbVie, NXE0033744 adds an investigational candidate with an EP4-mediated mechanism to its IBD development portfolio. Its potential differentiation will need to be evaluated through clinical studies demonstrating whether gut-selective EP4 activation can deliver meaningful and durable patient benefits.

Financial Terms and Expected Timeline

Nxera will receive US$100 million upfront and is eligible for up to US$1.37 billion in additional development and commercial milestone payments, plus tiered royalties on net sales. The milestone payments are contingent on achieving specified development and commercial events and do not represent guaranteed revenue.

The upfront payment is expected to be recognized as revenue in the fourth quarter of the financial year ending December 31, 2026, subject to completion of the transaction by December 31 and agreement with the group’s auditors. Nxera expects the payment to have a material positive impact on its revenue and financial position.

The companies have not disclosed timelines for subsequent clinical trials, regulatory submissions or potential commercialization. Completion of customary antitrust clearances is the immediate transaction-related requirement. Further development will depend on clinical outcomes and subsequent regulatory interactions.

Overall, the agreement provides Nxera with an upfront financial return and potential long-term royalties while placing NXE0033744’s global development under AbbVie’s leadership. For the candidate, the next critical step is to establish whether the pharmacological signals observed in Phase 1 can translate into clinical efficacy in patients with IBD.
